S. mutans Recombination (SMUR) Media Recipe:
Final Recipe:
For 45mL of SMUR Media:
- Add 30 mL of 1.5x SMUR Base
- Add 9.5 mL of Sterile MilliQ Water
- Add 4.5 mL of 10% Glucose
- Add 450 uL of 100x SMUR Metals
- Add 450 uL of 100x SMUR Unstables
- Add 90 uL of 500x SMUR Vitamins
- Vortex and store in 4˚C Fridge
Do not use for longer than 2 weeks, or if anything has precipitated.
Stock Solutions:
500mL 1.5x Base Media:
pH to 6.95 to 7.05, then autoclave.
45mL 100x Metals Solution:
Add MgCl2 last and make sure everything is dissolved before it is added. There will be an exothermic reaction and and color change upon addition of MgCl2. If there is trouble dissolving everything, heat water bath to 45˚C and heat solution while vortexing occasionally. Then filter sterilize.
40mL 500x Vitamins Solution:
If there is trouble dissolving, heat water bath to 45˚C and heat solution while vortexing occasionally. Then filter sterilize.
40mL 100x Unstables Solution:
If there is trouble dissolving, heat water bath to 37˚C and heat solution while vortexing occasionally. Then filter sterilize.
